Patented Optofluidic Force Induction (OF2i®)

Optofluidic Force Induction (OF2i®) is a non-destructive, parameter-free measuring method for particle counting, particle sizing and determination of particle concentration in liquid samples. Results are based on detection and counting of up to 1000 particles per minute in a continuous flow of sample through a measuring cell.

OF2i® measurement is:

  • Continuous & with single-particle sensitivity to monitor dynamic processes as they occur in the sample, e.g. aggregates forming, reactions, dissolution, etc.
  • Single-particle sensitivity is the basis for particle distributions which represent all the populations within polydisperse systems.
  • With capacity to measure large volumes OF2i® detects ultra-low particle concentrations as well as large-particle tails, LPC, anomalies and outliers.

OF2i® particle sizing is available with lab devices BRAVE B-Curious and PAT device BRAVE B-Continuous. It forms the basis for analysis with BRAVE B-Aware, BRAVE B-Phat and BRAVE B-Elementary.

Unique combination OF2i®-Raman

The combined OF2i®-Raman setup uniquely uses single-particle tracking to capture the scattered Raman signals of individual particles at up to 30 frames per second. The recorded Raman spectra are then compared with a reference spectrum, while the OF2i® signal calculates particle size based on the speed of each particle.

Additional large-particle detection for LPC and PFAT5 applications

BRAVE Analytics’ large-particle counting device, BRAVE B-Phat, detects large particles and aggregates from 2 µm to 80 µm in-flow and records their size based on dynamic imaging principles.

Large-particle detection is available with BRAVE B-Phat.

BRAVE devices in detail

The BRAVE B-Curious is a benchtop nanoparticle analyser designed for particle sizing, determination of particle size distributions and particle concentration in a size range from approx. 50 nanometers to 3 micrometers. With its continuous, time-resolved measurement, it excels in observing real-time changes in particle systems and capturing ultra-low concentrations quickly.

The BRAVE B-Aware module is an extension for the BRAVE B-Curious base station. It detects particle counts in a size range from approx. 5 to 50 nanometers. It is ideal for detecting the onset of aggregation and monitoring other changes in the particle count or concentration of small particles.

BRAVE B-Elementary is a Raman Spectrometry Module that can be added to the BRAVE B-Curious. It identifies substances, different particle populations, phases, and polymorphs in high-throughput screening. Its applications include detecting microplastics in water, non-targeted contamination screening, and quality control in ultra-pure water.

The BRAVE B-Continuous is a sensor for process analytics, particularly useful in pharmaceutical and biotech applications such as quality control of emulsions and for tracking changes in aggregation levels, for detecting contaminations, and ensuring the batch meets specifications without the delay inherent with lab analysis. It allows for scaling up laboratory results to production environments.

BRAVE B-Phat uses dynamic imaging techniques to detect large-diameter particles, particle tails and aggregates in the size range from 2 µm to 80 µm. It can be used for analysis of subvisible particles in protein therapy formulations, other injectables and emulsions, according to USP <729>, USP <788> and <USP 787>. There is a model for the laboratory and a PAT sensor version.

  • Monitoring the plant (including mixing, dilution, and filtering systems)
  • Gathering real-time data for process optimisation (data lake)
  • Quality control of water for injections

 

BRAVE B-Phat large-particle detector:

  • Continuous scanning for large-diameter particles
  • Therefore reliable determination of PFAT5 value acc. to <USP 729> during production of e.g. parenteral nutrition formulations
  • Also applicable for <USP 788> and <USP 787>.
  • Monitoring of contamination, anomalies and aggregates
